1. Sample Pretreatment and Loading Logic

The cell sample to be processed (typically a liquid suspension with a concentration adapted to the instrument requirements, generally 10 ⁶~10⁸ cells/mL) must first be mixed with an anticoagulant (e.g., EDTA, heparin) to prevent cell aggregation. Subsequently, the sample is injected into a specialized centrifuge tube (or sample well), and the glass slide is fixed onto the centrifuge’s specialized rotor body (such as the "specialized rotor body for cell smears" mentioned earlier). It is crucial to ensure the glass slide is parallel to the sample liquid surface with a uniform gap (usually 0.1~0.3mm) — this design serves as the foundation for the subsequent uniform spreading of cells, avoiding uneven smear thickness caused by gap variations.

2. Directional Driving Effect of Centrifugal Force

After the instrument is started, the rotor rotates at a set speed (common range: 200~2000 rpm, corresponding to a relative centrifugal force (RCF) of 50~720 xg, as per the previous parameter "2000rpm 720xg"), generating a radially outward centrifugal force. At this point, due to their significantly higher density than the liquid medium (e.g., plasma, culture medium), the cells in the sample overcome the viscous resistance of the liquid under the action of centrifugal force and undergo directional sedimentation toward the glass slide. The magnitude of the centrifugal force directly determines the cell sedimentation rate: an excessively low RCF results in incomplete cell sedimentation and sparse smears, while an excessively high RCF may cause cell lysis and morphological deformation. Therefore, parameters must be precisely matched according to the cell type (e.g., 1000~1500 rpm for blood cell smears and 800~1200 rpm for tumor cell smears).

3. Core Mechanism of Uniform Cell Spreading

When cells sediment onto the glass slide surface, the rotational movement of the rotor also generates tangential frictional force. Combined with the flow-guiding design of the sample well (wedge-shaped wells in some instruments), this force causes cells to form a gradient distribution (or uniform thin layer) from dense to sparse along the centrifugal direction on the glass slide. This process is analogous to "centrifugal force driving liquid flow to 'brush' cells onto the glass slide," ultimately forming a cell smear with a thickness of 1~5μm. The thickness can be adjusted by modifying the rotation speed and centrifugation time: higher speeds and longer durations result in denser smears, though cell stacking must be avoided.

4. Connection Between Termination and Sample Fixation

Upon reaching the set centrifugation time (typically 30~120 seconds; short-term centrifugation minimizes cell morphological damage), the instrument smoothly stops the rotor via its braking system (to prevent smear detachment or cell displacement due to inertia). At this stage, the cells have been fixed to the glass slide surface through physical adsorption and can be directly removed for subsequent processing (e.g., methanol fixation, HE staining, immunohistochemical staining). Finally, the smear is used for pathological diagnosis, cell morphological analysis, or scientific research observations.

Adaptability Between Key Designs and Principles

They ensure centrifugal force consistency across all stations (error ≤ ±5%), guaranteeing the uniformity of batch-prepared smears.
  • Precise Control of Rotation Speed and RCF: The instrument adjusts the rotation speed via a variable-frequency motor to stabilize the RCF within the target range, avoiding variations in cell smear quality caused by centrifugal force fluctuations.
  • Contamination Prevention Design: The parts of the rotor in contact with samples are made of inert materials (e.g., stainless steel, PTFE coating), and the sample wells are independently separated to prevent cross-contamination, meeting the sterility requirements of clinical testing.
