Molecular hybridization analyzer, also known as molecular hybridization furnace or molecular hybridization box, is a device that uses nucleic acid molecular hybridization technology to detect whether the tested genome contains known gene sequences. Widely used in gene cloning screening, enzyme digestion mapping, detection of specific gene sequences in the genome, and genetic diagnosis of genetic diseases.

Principle of Molecular Hybridizer:

Nucleic acid hybridization technology, based on the principle of complementary base pairing, uses labeled known DNA or RNA fragments (probes) to detect the nucleic acid sequence to be tested in a sample under certain conditions. Under physiological conditions, DNA exhibits a stable double helix structure. In vitro, when there are denaturing factors present (such as temperature exceeding 65 ℃, amine content, or terminal pH), the hydrogen bonds inside the DNA molecule break and dissociate into two single stranded DNA. This phenomenon is called denaturation. After removing denaturation factors, single stranded DNA can recombine through base complementarity to form a stable double helix structure, a process known as refolding or annealing. In the process of refolding, if there are exogenous single stranded DNA fragments or oligonucleotides (usually 17-45 bases) that are identical or highly homologous to a certain part of the sample nucleic acid sequence, the two can form a hybrid through complementary bases, that is, a double stranded DNA molecule. This process is called hybridization. If a known DNA fragment is added to the refolding DNA and can bind to a double stranded molecule, it indicates that the sample contains a known DNA sequence.
